The CRTP is an outward-facing, multi-disciplinary hub purposed to enable the external cancer research community. CRTP has two major focus areas, the NCI RAS Initiative with the goal to discover new therapeutic interventions against RAS-driven cancers and to provide CCR and DCEG with multidisciplinary technology expertise needed for address the complex etiology of human cancer. In addition, the CRTP hosts the Nanotechnology Characterization Laboratory (NCL) which performs and standardizes preclinical efficacy and toxicity testing of nanoparticles intended for cancer therapeutics and diagnostics, and the Antibody Characterization Lab (ACL) which characterizes monoclonal antibodies or other renewable affinity binding reagents for use in cancer related research.

KEY ROLES/RESPONSIBILITIES

The Protein Characterization Laboratory (PCL) provides support to the NCI CCR and DCEG in areas of protein characterization and proteomics, macromolecular interaction and targeted and global metabolite analysis using variety of separation techniques and state of the art mass spectrometry.

The Post-Baccalaureate Fellow will:

- Develop new technologies for improved identification of HLA peptides, crucial for cancer vaccine development
- Perform transient transfection in tissue culture cell to evaluate HLA peptide generation following over expression of specific target proteins
- Generate stable cell lines, over expressing both wild type and mutant form of specific test proteins, following evaluation of HLA peptide isolation efficiency
- Perform large scale purification of HLA bound peptides and develop methods to improve immunoprecipitation of HLA receptors
- Large scale purification of HLA antibodies for immunoprecipitation
- Work with senior scientist to systematically explore methods to improve ionization of HLA peptides
- Acquire mass spectrometry data following enhanced ionization and affinity purification of HLA peptides
- Raw mass spectrometry data processing using novel data processing tools
- Provide timely updates of project progress to supervisor and other staff in both informal and formal reports
- Maintain detailed records of all laboratory processes and procedures for quality assurance purposes
- Work in a team-oriented environment to carry out technology development efforts The position is funded for a period of one year.
